ABSTRACT

Objective@#To explore the relationships among social adaptation, physical exercise, and mental health of adolescents, so as to provide theoretical basis for social adaptation and mental health improvement.@*Methods@#Longitudinal follow up survey data were collected from 1 163 adolescents in Jinan City, Shandong Province in September 2021 (T1), January 2022 (T2), and June 2022 (T3) by cluster random sampling method. The relationship between social adaptability, physical exercise, and mental health of adolescents was analyzed through cross lagged analysis.@*Results@#There were significant sex differences in physical exercise and mental health among adolescents on T1, T2, and T3 ( Z =-3.83, -3.43, -4.59; -12.45, -8.93, -8.72, P <0.01), with male students had more physical exercise [35(28,42),36(33,42),38(35,43)] and better mental health [12(12,17),17(17,21),14(14,26)] levels than female students [33(27,40),35(31,40),36(33,41); 9(9,12),12(12,23),12(12,23)]. No similar sex difference in social adaptability (T1, T2, T3) [male: 22 (14, 24), 22 (14, 24), 22 (16, 24); female: 21 (11, 23), 20 (14, 24), 22 (17, 24)] ( Z =-0.79, -1.19, -1.34, P >0.05). Temporal and synchronous correlations were found in social adaptability, physical exercise and mental health ( r =0.18-0.67,0.12-0.68, P <0.01). Teenager physical exercise and mental health could predict social adaptability across time ( β T1-T2 =0.25,0.13; β T2-T3 =0.25,0.06, P <0.05), with a mediating effect on the impact path of physical exercise on social adaptability in psychological health ( β =0.14, P <0.05).@*Conclusions@#There is a causal relationship between physical exercise, mental health, and social adaptability among adolescents. Encouraging adolescents to actively participate in physical exercise and promoting mental health can effectively enhance their social adaptability.

ABSTRACT

Objective@#To analyze the changes in physical health among primary and secondary school students in Jinan during 2019 to 2022, so as to provide reference for primary and secondary school students physical health promotion.@*Methods@#From 2019 to 2022,16 496 primary and junior middle school students will be selected from Jinan by cluster random sampling method, the comprehensive scores of physical health for each year were calculated according to the National Student Physical Health Standards (revised in 2014), and changes in physical health were analyzed using t-tests, univariate analysis, analysis of covariance and χ 2 tests.@*Results@#The students physical health scores ranged from high to low in 2022(80.78±12.63),2021(79.12±12.72),2019 (75.17±2.03)and 2020(73.82±12.63). During 2019 to 2022, the overall physical health score of female students were higher than that of male students ( F=105.39, 288.29, 197.90, 159.36, P <0.01). The differences in the comprehensive physical and health scores of male and female students in different grades from 2019 to 2022 were statistically significant ( F=34.22, 34.96, 20.33, 45.08 , 13.56, 18.29, 24.86, 30.01, P <0.01). The individual indicators showed varying degrees of decline in 2020. Except for body mass index (BMI), the fluctuations in lung s capacity, run 50 meters, sit forward, standing long jump, 1 minute sit-ups, pull-up, endurance running, 1 minute skipping rope during 2019 to 2022 were statistically significant (primary school: F =127.58, 44.80, 223.64, 76.66, 10.69, 296.79, secondary school: F =189.31, 52.10, 63.12, 61.49, 7.34, 8.10, 50.94 , P <0.01). The detection rates of overweight and obesity in primary and secondary school students from 2019 to 2022 were 30.09 %, 31.45%, 30.14 %, and 30.60%, respectively ( χ 2=2.26, P >0.05). From 2019 to 2022, the rates of overweight and obesity in primary and middle school boys were 34.56 %, 34.66%, 32.14%, 33.73%, 37.41%, 43.53%, 41.36 %, 39.16%, respectively, and the rates of overweight and obesity in primary and middle school girls were 24.66%, 24.39%, 24.61%, 24.65%, 23.82%, 28.74%, 26.99 %, 27.95%, respectively, none of which were statistically significant ( χ 2=2.93, 4.29 , 0.04, 3.12, P >0.05).@*Conclusion@#During 2019 to 2022, there are no significant changes in physical health indicators among primary and secondary school students in Jinan, and the overall level of physical health recovered rapidly after a slight decline due to the COVID-19 epidemic.

ABSTRACT

@#To construct PTEN/PLGA-(HE)10-MAP nanoparticles, which encapsulated PTEN plasmid DNA and combined with the pH-responsive cell-penetrating peptides (CPPs), and to investigate their effects of gene delivery and anti-tumor targets in vitro. Poly (lactic-co-glycolic acid) (PLGA) nanoparticles loaded with PTEN plasmid DNA were prepared by double emulsification-solvent evaporation method. PTEN/PLGA-(HE)10-MAP nanoparticles were prepared by coupling the histidine-glutamic acid-model amphipathic peptide nanocomplex [(HE)10-MAP] to the surface through amide condensation reaction. Particle size, Zeta potential, encapsulation rate and drug loading were tested to characterize the nanoparticles. By analyzing the cytotoxicity, cellular uptake, targeted transfection of eukaryotic expression plasmids and anti-tumor cell proliferation, the feasibility as a targeted gene delivery system were evaluated. The particle size of PTEN/PLGA-(HE)10-MAP nanoparticles was (266.5 ± 2.86) nm, with the encapsulation efficiency (80.6 ± 6.11)%. Zeta potentials were -(6.7 ± 0.26) mV, +(0.7 ± 0.22) mV and +(37.5 ± 0.85) mV at pH 7.4, 7.0 and 6.5, respectively. In the cytotoxicity test, the cell survival rates of tumor and normal cells were above 80%.Non-loading PLGA-(HE)10-MAP nanoparticles showed no obvious cytotoxicity. The results of cellular uptake experiments showed that PTEN/PLGA-(HE)10-MAP nanoparticles were more readily taken up by cells.The results of CCK-8 showed that the nanoparticles could pH-specifically inhibit proliferation of tumor cell in vitro.And PTEN/PLGA-(HE)10-MAP nanoparticles may be applied in tumor gene therapy.

Background@#Sleep disorders are one of the earliest non-motor symptoms of Parkinson’s disease (PD). Sleep disorders could, therefore, have value for recognition and diagnosis in PD. However, no unified classification and diagnostic criteria exist to evaluate sleep disorders by polysomnography (PSG). Utilizing PSG to monitor sleep processes of patients with PD and analyze sleep disorder characteristics and their relationship with demographic parameters could aid in bridging this gap. This preliminary study aimed to evaluate the clinical characteristic of sleep disorders in PD using PSG.@*Methods@#PSG was used to evaluate sleep disorders in 27 patients with PD and 20 healthy volunteers between August 2015 and July 2018 in Fujian Medical University Union Hospital. Total sleep time (TST), sleep efficiency (SE), total wake time, and other parameters were compared between the two groups. Finally, the correlation between sleep disorders and age, disease duration, Unified Parkinson’s Disease Rating Scale-III scores, Hoehn-Yahr stage, and levodopa dose were analyzed. The main statistical methods included Chi-square test, two independent samples t test, Fisher exact test, and Pearson correlation.@*Results@#Sleep fragmentation in the PD group was significantly increased (74.1%) while difficulty falling asleep and early awakening were not, as compared to healthy controls. No significant differences were found in time in bed, sleep latency (SL), non-rapid eye movement (NREM) stage 1 (N1), N1%, N2, N2%, N3%, and NREM% between PD and control groups; but TST (327.96 ± 105.26 min vs. 414.67 ± 78.31 min, P = 0.003), SE (63.26% ± 14.83% vs. 76.8% ± 11.57%, P = 0.001), R N3 (20.00 [39.00] min vs. 61.50 [48.87] min, P = 0.001), NREM (262.59 ± 91.20 min vs. 337.17 ± 63.47 min, P = 0.003), rapid-eyemovement (REM) (32.50 [33.00] min vs. 85.25 [32.12] min, P < 0.001), REM% (9.56 ± 6.01 vs. 15.50 ± 4.81, P = 0.001), REM sleep latency (157.89 ± 99.04 min vs. 103.47 ± 71.70 min, P = 0.034) were significantly reduced in PD group.@*Conclusion@#This preliminary study supported that sleep fragmentation was an important clinical characteristic of sleep disorders in PD. Whether sleep fragmentation is a potential quantifiable marker in PD needs to be further investigated in the future study.

ABSTRACT

Objective To develop an ECG monitoring and management system based on the smartphone platform to transmit ECG data to the doctor in daily life or working environment.Methods The system was composed of an ECG signal acquisition device and a smartphone App.The ECG signal acquisition device used MSP430F149 microcontroller as the core unit,which executed ECG signal extraction by adopting right leg drive,active filters,dual instrumentation amplifier and moving average algorithm,applied differential threshold method to accurately detecting R wave,calculated heart rate and then sent ECG data to the smartphone through Bluetooth 4.0 module.The smartphone App was developed based on cross-platform mobile development framework Cordova,and used Cordova plugins,JavaScript,HTML5 and CSS to receive the data from the acquisition device,draw and store ECG,show heart rate and remind abnormalities in heart rate as well as to transmit ECG data to the doctor through the mobile medicine App and social App.Then the doctor provided advices on prevention,treatment and etc after analyzing the received data.Results Compared with the ECG machine,the system had the relative errors of heart rate,P duration,QRS duration and QT duration being 2.7%,4.5%,4.3% and 3.3% respectively.All the above four parameters having significant correlations,and the correlation parameters were 0.955,0.948,0.91 and 0.834 respectively (P<0.05).The system had 8-h endurance in case of being fully charged.Concluslon The system gains advantages in power consumption,accuracy,noise,cost,volume,weight,safety,reliability as well as easy operation and maintenance,and facilitates the ECG conditions to be mastered timely by the doctor.

ABSTRACT

OBJECTIVE: To determine whether curcumin reverses the multidrug resistance of human colon cancer cells in vitro and in vivo. METHODS: In a vincristine-resistant cell line of human colon cancer, the cell viability of curcumin-treated cells was determined using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. Rhodamine123 efflux was evaluated to detect P-glycoprotein transporter activity, and expression of the multidrug resistance protein 1 and survivin genes was analyzed by reverse transcription polymerase chain reaction and western blotting. In addition, xenograft mouse tumors were grown and treated with curcumin. The morphology of the xenografts was investigated by hematoxylin-eosin staining. The in vivo expression of the multidrug resistance gene and P-glycoprotein and survivin genes and proteins was observed using reverse transcription-polymerase chain reaction and western blotting, respectively. RESULTS: Curcumin was not obviously toxic to the vincristine-resistant human colon cancer cells at concentrations less than 25 μM, but the growth of cells was significantly inhibited. At concentrations greater than 25 μM, curcumin was toxic in a concentration-dependent manner. The sensitivity of cells to vincristine, cisplatin, fluorouracil, and hydroxycamptothecin was enhanced, intracellular Rhodamine123 accumulation was increased (p<0.05), and the expression of the multidrug resistance gene and P-glycoprotein were significantly suppressed (p<0.05). The combination of curcumin and vincristine significantly inhibited xenograft growth. The expression of the multidrug resistance protein 1 and survivin genes was significantly reduced in xenografts of curcumin-treated mice and mice treated with both curcumin and vincristine relative to control mice. CONCLUSION: Curcumin has strong reversal effects on the multidrug resistance of human colon carcinoma in vitro and in vivo. .

ABSTRACT

<p><b>OBJECTIVE</b>To investigate chloride channel 1 (CLCN1) gene mutation and clinical features of 2 Chinese patients with myotonia congenita.</p><p><b>METHODS</b>Clinical data of a patient from a family affected with myotonia congenita in addition with a sporadic patient from Fujian province were analyzed. Exons of CLCN1 gene were amplified and sequenced.</p><p><b>RESULTS</b>The proband from the affected family was found to carry a c.1024G>A heterozygous missense mutation in exon 8, whilst the sporadic patient has carried a c.1292C>T heterozygous missense mutation in exon 11.</p><p><b>CONCLUSION</b>Detection of CLCN1 gene mutation is an effective method for the diagnosis of myotonia congenita. Exon 8 of CLCN1 gene may be a mutational hotspot in Chinese patients with myotonia congenita.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the possible effects and roles of bodyweight on the puberty onset in adolescent girls.</p><p><b>METHODS</b>Totally 288 Chinese female children and adolescent girls aged 5 to 16 were followed up yearly for four consecutive years. The height, bodyweight, fat percentage, sexual characteristics, and the serum levels of leptin and insulin-like growth factor-1 (IGF-1) were studied to analyze the influential factors of puberty onset and age of menarche.</p><p><b>RESULTS</b>The serum level of leptin elevated significantly from age 13 [(9.23 +/- 1.25) microg/L] and reached peak at age 16 [(13.19 +/- 1.45) microg/L]. IGF-1 significantly correlated with the timing of puberty onset (r = 0.292, P = 0.016). BMI and fat percentage had no significant effects on the onset of puberty, but were negatively correlated with the age of menarche (r = -0.323, P = 0.037, r = -0.298, P = 0.038 respectively).</p><p><b>CONCLUSION</b>Bodyweight may have effect on puberty onset in female adolescents.</p>

ABSTRACT

This study was aimed to investigate the serologic characteristics, genetic background and population distribution of B2 and AB2 subtype in Chinese ABO blood group. The classic blood group serological technology was used to detect ABO blood group of the propositus and their family members, the anti-B1 serum prepared by yourself was used to investigate the distribution of B1/B2 and AB1/AB2 subtype of the blood donor. The results indicated that the antigen of propositus was AB2 subtype and that of his child was B2 subtype. The anti-B1 antibody was detected in blood serum of propositus; the antigen of 3 from 2318 blood donors with B blood group were found to be B2 subtype, the antigen of 2 from 826 blood donors with AB blood group were found to be AB2 subtype. The investigation on propositus and the 3 B2 blood donor families showed that B2 antigen displays genetic characteristics of blood group. It is concluded that B2/AB2 subtype is from family inheritance, while B2 subtype is amounted to 0.129% in B blood group, and AB2 subtype is amounted to 0.224% in AB blood group.

ABSTRACT

Objective To further investigated the effect of minocycline on the inhibition of microglial activation and subsequent protection of nigral DA neuron.Methods 20 rats injected with LPS in the substantia nigra (SN) were randomly divided into two groups (LPS group and LPS+Minocycline group).The behavior was observed on the 7~(th) d and 14~(th) d.The immunohistoehemistry,in situ hybridization and Western-blot were used to detect the levels of positive neuron,mRNA,protein of TH and OX-42. Results The slightly rotational behavior was observed in LPS+Minoeyeline group.The majority of mieroglias were activated in the two groups.Some microglia in the SNpc remained ramified in LPS+ Minocycline group.The numbers of hypertophie microglia in LPS+Minoeyeline group were less than that in LPS group.Western-blot showed that the protein of OX-42 in two LPS groups was higher than in normal group(P
